In all honesty, it's a judgment call.
With comics like Hellboy, or anything that's on Dark Horse, there's a fixed "This is the first book" kind of deal since it's a relatively new comic.
But yeah, with things like X-Men and Spiderman, they've just been around so long they have to continually rehash. I wouldn't bother starting from the beginning of the story, I'd start at the beginning of a certain arc, like Ultimate X-Men.
For you though, X-Men I'd say Uncanny X-Men. Picks up after the 3rd film with elements of origins as well, and deals with the rebirth of the Phoenix force clone.
Deadpool, go for Deadpool Volume 1 (nice and easy

Spidey, Amazing Spider-Man: Volume 1: Coming Home has the best artwork, and great characterization.
I'm insanely picky with what I'll read, if the artwork isn't so amazing I think my brain might melt, or the storytelling isn't absolutely breathtaking, it'll be either in the bin, or back to the shop before I've got past the 2nd chapter
Khei: Graphic novels are just comics that were released as single issues, sown together in to a volume for convenience, so you're still just reading comics.
